Is there a way that I can define hotkeys for my brushes? So if I press the key, the wanted brush is choosen? This would speed up painting a lot...
perhaps also a plugin or something like that?

Hi,
No hotkeys/keyboard shortcuts for brush variants.
Use either Custom Palettes or the Tracker palette to access most used brush variants.
Using Custom Palettes, you can click an icon to access the brush category and the brush variant selected when the icon was added to the Custom Palette. The icon can also be clicked to load the library in which that brush category and brush variant are located.
You'll find information on Custom Palettes in the following locations (not available in Painter 8, though available with the Painter 8.1 patch installed):
Painter IX Help > Help Topics > Contents tab > A Workspace Tour
On the right panel, click Custom Palettes
