The usual suspect for this is the Wacom driver going buggy. You can try several things:

First off you can try just removing your Preference File from the Wacom Tablet Preference File Utility and see if that helps? Try removing both the choices ie My Preferences and All User Preferences. For me, one did absolutely nothing and the other worked, but I can't remember which it was. Sometime reinstalling the wacom driver does not remove this info so it still causes problems if it develops a glitch.

If that doesn't work uninstall the driver completely, shutdown and restart your system, then reinstall. Try it with the newest driver (download from wacom). If you still experience problems, try an older driver. Remember, it is best to uninstall and reboot, then reinstall the Wacom drivers, especially if you are having problems. They are notoriously temperamental and a reinstall works best if you have completely cleared your system of the old driver.

This was happening to me before updated of wacom...quite a while back. Make sure the Wacom Driver is current and that Painter is up to date as well.
