I'm guessing you're using it on Windows? I tried to use it a while back out of curiosity... my tablet would stop responding randomly, but the mouse would work; then both the mouse and tablet would stop responding on the canvas.
Maybe I was doing it wrong or something, but I hate it. It's terribly unintuitive. I assume it's called The Gimp for a reason.

I have gimp; I can make marks with it, but not very good ones. I fail! The bad workman blames his tools; so I'm not going there.
Does sound like something's wrong, though. I'd suggest you uninstall and try again....

if it is a windows version, try to install the last stable version(2.6) or the last stable bug fix version(2.6.3). you can tweak configuration of devices in the preference menu, however this is not necesary on windows.
if you have a linux version, check the xorg configuration to see if you have a correct tablet configuration. if the xorg file is configured(see the linux wacom project page for detailed information) then you need to configure the input devices on gimp(edit/preferences/input devices) put the stylus on "screen" and the same for the eraser.
in any case more information of your problem could help(version, os, pen panel options)
good luck

Preface: I'm not trying to be a smartass here. You never know.
Have you looked at your task bars to make sure you are on brush instead of eraser? Have you zoomed in deeply to see if the thing is making marks so small you can't see them? Make sure your opacity is not set to zero. Are you accidentally stuck in "select rectangle" (you would see dashed lines following your pen around). Did you try to draw a little something in Paint (windows) or Kolourpaint (linux). just to make sure your tablet is working?
If you are in linux, what distribution are you using? Debian stuff like the Xandros OS might not readily accept add ons that come in certain file types. I think it's .tbz that's the safest (I have to go back and read my notes to see if I got that right). Even then you have to compile it carefully and some other bits of code are required to help you do that. It's easiest when you got a packaged distribution to go get your GIMP from those guys rather than from the gimp.org or its mirrors. Xandros gave me a version that works like a charm and installed itself into my Xandros OS. No fuss, no muss.
