Just find a comfortable position and start drawing. I'm not sure how big the Bamboo's are, but if it's anything like a 6x8 tablet, the way that I have it is so that the bottom rests on my chair's armrest, and the top is pushing against the edge of my best. Lets me use my elbow is a pivoting point as well as give me a comfortable angle to work at.
An exercise that I read about is just picking a large brush and making a series of concentric rings, then choosing a smaller brush and trying to follow the same path inside of it. This just helps you get the feel of how your pen is gonna work in correspondence to the computer screen.
As far as settings go, those are all by both preference & need. I personally just use the opacity jitter in Other Dynamics. But sometimes it's good to play around with flow jitters, spacing jitters, size jitters, etc. Just open up a blank canvas and start playing around with stuff.
It's a completely different feeling than drawing with a pencil. People have this conception that getting a tablet will let them draw exactly as they would with a physical medium. To an extent this is correct, but it's just like being someone who draws with graphite learning how to use oils. It's just something you have to practice with and learn to do.

I found this tutorial (http://www.dustyghost.com/tablet_tut.html) to be outstanding when I first got my graphics tablet!!

Another thing is to go into the main settings on the tablet (in control panel -pc/system preferences -mac) & adjust them to where it is used to the amount of pressure you're comfortable with in regards to using the pen. In Painter there's a section in the preferences on gauging this. Just play around with the settings & keep going back into Photoshop to try things out, once you get it set to how you work it will speed things up quite a lot from just using a mouse.
